白細胞介素15(IL-15)是一種很異常的細胞因子,它和IL-2具有很多相同的生物學特性,它最先在猴腎上皮細胞系(CVI/EBNA)的培養(yǎng)條件下被發(fā)現(xiàn),其可促進小鼠T細胞系(細胞毒性T淋巴細胞株)的有絲分裂活動。IL-15和IL-2具有相似的生物活性包括刺激自然殺傷細胞、 激活的外周血T淋巴細胞、 腫瘤浸潤淋巴細胞和B細胞的生長。IL-15也是人血液中T淋巴細胞的趨化因子,也可誘導NK細胞中淋巴因子激活的殺傷活性和誘導殺傷效應細胞的產(chǎn)生。另外,IL-15對不參與免疫反應的細胞也有影響。 骨骼肌細胞不僅可以表達IL-15和IL-15受體mRNAs.也對IL-15也有反應。人的IL-15和猴子、小鼠的IL-15分別具有97%和73%的氨基酸序列同源性。人和猴子的IL-15都可作用于小鼠細胞。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法??谷薎L-15單抗包被于酶標板上,同時加入標本、標準品和生物素化的抗人IL-15抗體,標本、標準品中的IL-15會與加入于孔中的生物素化的抗人IL-15 抗體及包被于酶標板上的單抗結合,形成免疫復合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結合,游離的成分被洗去。加入顯色底物(顯色劑),若反應孔中有IL-15,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值,IL-15濃度與OD450值之間呈正相關,可通過繪制標準曲線求出標本中IL-15濃度。
